The candidate will optimize an instrumentation and a microfluidic card for encapsulating human cells in monodisperse microbeads of extracellular matrix. Within a few days, cells encapsulated in microbeads form organoids or tumoroids, which are miniature 3D cellular models of biological tissues or tumors. Based on the microfluidic instruments previously developed in the team, you will design and assemble an encapsulation instrument incorporating functions for automatic counting and transfer of microbeads to the appropriate medium for organoid and tumoroid culture. Your mission will be to adapt the team's previous work and develop a new high-throughput cell encapsulation instrument comprising a microfluidic card, a mechanical holder, pneumatic controllers, temperature control units, optical systems and IT tools. These technical developments will be carried out and validated in collaboration with biologist users. Required work: - Analysis of requirements and definition of technical specifications to meet them: scale-up + GMP production of organoids and tumoroids - Design of the optimized functional prototype - Incorporation of microbead counting and transfer functions - Validation and qualification of the bioproduction instrument - Design specifications of the pre-industrial prototype Activities: - Instrumentation / interfacing - Dimensioning of the components - CAD design (SolidWorks) of the instrument including fluid management, thermal management, fluid management (liquids and gases), imaging systems, image processing module. - Design and production of parts by 3D printing - Management of subcontracting: supply of components, machining - Parameterization of components - Assembly of the equipment - Functional validation of the developed equipment
